study,aimed to determine,the involvement of long non-coding RNA PVT1 in the progression of cardiac hypertrophy.,Methods,A C57BL/6 mouse cardiac hypertrophic model,was established using transverse aortic constriction(TAC)operation.,Expression level of PVT1,was analyzed using quantitative real-time PCR(qRT-PCR)and outcome indicators for cardiac hypertrophy were detected by echocardiography analysis after TAC operation at 4 weeks.To further investigate the roles of PVT1 in cardiac hypertrophy,PVT1 siRNA(siPVT1),was transfected into cultured neonatal mouse cardiac cardiomyocytes which was subjected to,1 M angiotensin II(Ang II)stimulation,for 24 h.,Cell size,was calculated by measuring cell surface area and,total protein content,was analyzed by Bradford assay.Moreover,the,expression levels of hypertrophic markers,including atrial natriuretic peptide(ANP),B-type natriuretic peptide(BNP)and beta-myosin heavy chain(-MHC),were also quantified using qRT-PCR.,Results,Compared with the sham-operated group,PVT1 was up-regulated by,2.5-fold(,p,0.05,),in hypertrophic hearts after TAC operation,at 4 weeks,.SiPVT1 transfection caused,significant decrease(,p,0.05),of Ang II-induced enlarged cell size by 5.6-fold and increased total protein content,by 23.0%.,Moreover,siRNA transfection,dramatically(,p,0.05)attenuated,Ang II-induced up-regulation of ANP and-MHC,by 40.9%and 41.5%,respectively,but had no effect on BNP expression.,Conclusions,Our results,demonstrated that,PVT1,was essential for,the maintenance of cell size of cardiomyocytes,which,might play a ro,le,in,the failure via affecting mitochondrial oxidative stress 29.Cell enlargement,a feature of cardiomyocyte hypertrophy,is associated with heart systolic dysfunction and heart failure 30.Total protein content of cardiac muscle cells usually increases significantly in cardiacmyocyte hypertrophy 31.Herein,we observed an enlarged cell size and increased total protein content in Ang II-treated cardiomyocytes,implying an successful establishment of cardiacmyocyte hypertrophy in vitro,.At the same time,PVT1 knockdown could significantly attenuate the increase of cell size and total protein content in Ang II-treated cardiomyocytes,which give the evidence that,PVT1 could be suggested as a therapeutic targets for disease treatment of cardiac
